Daganattalálás carcinoem bryonális antigén-expresszáló daganaton, transzgén egérben

ÖSSZEFOGLALÁS:

Carcinoembryonális (CEA) gént hordozó egérben vizsgáltuk rádióizotóppal jelzett monoklonális antitestekkel (Mab) a biológiai megoszlást (biodistributio) és tumor találást. A tumorban mért specifikus antitest felvételt mértük CEA-transzfektált, syngén daganatot hordozó egérben és antigén-negatív, eredeti, ugyanezen típusú tumorban. A magas CEA-expresszáló syngén tumorban az antitest beadott dózis %/gramm (%ID/g) szövetben kifejezett felvétel, 48 órával a beadás után elérte a 30%-ot. Az antigén-pozitív tumorban a dúsulás 5-8x volt magasabb, mint az ugyanabba az egyedbe beültetett, antigénnegatív, eredeti daganatban. Immunoscintigráfiával csak az antigén pozitív daganatok váltak láthatóvá. A humán xenograft-daganatot hordozó tímuszmentes csupasz egérben nyert tumor találás hasonló volt, mint a CEA-expresszáló egér-daganatot növelő-transzgén-, és a tímuszmentes csupasz egérben is. A daganat lokalizáció mértéke a CEA-traszfektált daganatokban az antigén expresszió mértékétől függött. Keringő immun komplexek nem voltak. Az antitest kiürülése az érpályából hasonló volt transzgén és nem transzgén egérben. Kivételt a vastagbél képezett. Más normál szövetben a jód izotóppal jelölt monoklonális antitest megoszlása hasonló volt CEA-transzgén-, és nem-transzgén egérben. A vastagbélben a fokozott halmozódás nagyobb antigén pozitivitást sugall. Az eredmények a CEA-transzgén egér modell hasznosságát mutatják az antitestek kifejlesztéséhez, radioimmunodetektálásra és a CEA-expresszáló daganatok kezelésére.

SUMMARY

Mice transgenic for the carcinoembryonic (CEA) gene were used to study the biodistribution and tumor targeting of a radioiodinated monoclonal antibody (MAb), T84.66. The specificity of antibody uptake in tumors was assessed in mice bearing a CEA-transfected syngeneic tumor as well as the antigen-negative parental tumor. With high CEA-expressing tumors, the percent injected dose per gram (%ID/g) approached 30% at 48 hr. Tumor uptake in antigen-positive tumors was 5-8-fold higher than that observed in the antigen-negative parental tumors. Only antigen-positive tumors were visualized by immunoscintigraphy. The tumor targeting obtained in athymic nude mice bearing human tumor xenografts was similar to that observed with CEAexpressing murine tumors implanted in either athymic nude or transgenic mice. The degree of localization of CEA-transfected murine tumors was related with the level of antigen expression. Circulating antigen-radioantibody complexes were not detected while blood clearance of radio-antibody was similar between transgenic and non-transgenic mice. With the exception of the large bowel, the distribution of radioiodinated MAb in normal tissues was similar in both CEA transgenic and nontransgenic mice. Increased localization of intact antibody was observed in the large bowel from transgenic mice, suggesting specific targeting to antigen-positive normal tissues. These results suggest that the CEA transgenic mouse model will be useful in the development of antibodies for radio-immunodetection and treatment of carcinomas expressing CEA.
